Methylene Blue

Methylene Blue

3,7-bis(dimethylamino)phenothiazin-5-ium chloride

A synthetic compound with over 130 years of medical history — originally developed as a textile dye and later adopted as one of the first modern pharmaceuticals. At the low doses used in nootropic formulations, Methylene Blue is emerging as a promising cognitive enhancer that supports mitochondrial energy production, neuroprotection, and mental performance through unique mechanisms not found in other supplements.

Benefits: Energy, Focus, Neuroprotection
Type: Other
Forms: Capsule, Liquid

What Is It?

Methylene Blue (MB) was first synthesized in 1876 and has one of the longest medical histories
of any compound used today. It has been used as a treatment for methemoglobinemia, a malaria
treatment, a surgical dye, and is on the World Health Organization's List of Essential Medicines.

In recent years, neuroscientists and longevity researchers have turned their attention to
Methylene Blue at micro-doses (1-10mg) for its remarkable effects on mitochondrial function,
brain energy metabolism, and neuroprotection — creating significant interest in its potential
as a cognitive performance compound.

How It Works

Methylene Blue works through several unique mechanisms:

- Mitochondrial electron carrier — MB can accept and donate electrons directly in the mitochondrial electron transport chain, acting as an alternative electron carrier that improves cellular energy (ATP) production efficiency — even when the normal chain is compromised

- Cytochrome c oxidase enhancement — increases activity of Complex IV in the mitochondrial chain, the rate-limiting step in cellular energy production

- Antioxidant cycling — MB cycles between oxidized (blue) and reduced (colorless) states, neutralizing reactive oxygen species and reducing oxidative stress in brain tissue

- Cerebral blood flow — research suggests MB increases regional cerebral blood flow, improving oxygen and glucose delivery to brain tissue

- Neuroprotection — has demonstrated protective effects against tau aggregation and amyloid formation
in laboratory research

Side Effects & Considerations

Methylene Blue has an extensive safety record at medical doses, with important notes for
nootropic use:

- Will temporarily turn urine blue or green — this is completely harmless and expected
- May temporarily tint skin or mucous membranes blue at higher doses
- IMPORTANT: Do NOT combine with serotonergic medications including:
- SSRIs or SNRIs
- MAO inhibitors
- Tramadol or other serotonergic drugs (Risk of serotonin syndrome even at low doses)
- Do not combine with Kanna/Sceletium extracts for the same reason
- Not for use during pregnancy or nursing
- Those with G6PD deficiency should not use Methylene Blue
- Consult your healthcare provider before use if on any medications
- The 5-10mg doses in Natty Addy are within the nootropic dose range — stay within recommended serving guidelines

Typical Dosage Range

1mg – 10mg daily

The nootropic dose range for Methylene Blue is 1-10mg — significantly lower than pharmaceutical doses used medically. Research suggests a hormetic dose response where low doses are beneficial and higher doses may be counterproductive. Natty Addy provides 5mg (tablet) or 10mg (shot) — stay within these guidelines and do not exceed recommended servings. Best taken earlier in the day to align with
natural energy rhythms.

Research Notes

- Tucker et al. (2018) — Randomized controlled trial published in Psychopharmacology found low-dose Methylene Blue significantly improved sustained attention and short-term memory in healthy adults.

- Atamna & Kumar (2010) — Research in Aging Cell established Methylene Blue's mechanism as a mitochondrial electron carrier and its neuroprotective effects against age-related mitochondrial decline.

- Bhatt et al. (2021) — Review in Neurochemical Research documented Methylene Blue's emerging role in
cognitive enhancement and its potential therapeutic applications for neurodegenerative conditions.
